Study on the stability of SARS-CoV-2 at different temperatures

    目的观察不同温度保存条件下细胞培养物中新型冠状病毒(简称“新冠病毒”)存活情况,判断温度对病毒稳定性的影响,为新冠病毒肺炎疫情趋势研判及防控提供基础数据。
    方法将病毒接种于Vero E6细胞适应培养后,收获病毒液,根据所测得病毒半数组织培养感染剂量(TCID50)将不同稀释度(10-1、10-3、10-5、10-6)的病毒在不同温度下(4 ℃、22.5 ℃、37 ℃)保存1~7 d,并分别感染细胞,通过观察细胞病变效应(CPE)、实时荧光定量检测病毒核酸确定病毒感染性,以评价病毒在不同温度条件下的稳定性。
    结果不同浓度的新冠病毒在4 ℃条件下保存较为稳定,均具有感染性;22.5 ℃条件下,高浓度(10-1稀释度)病毒放置7 d感染性逐渐下降,其他较低浓度病毒放置1 d则完全失去感染性;37 ℃保存超过1 d病毒即失去感染性。
    结论在细胞培养环境中,新冠病毒在4 ℃条件下高度稳定,对热敏感,且与病毒浓度相关,高浓度病毒室温22.5 ℃条件下仍可存活7 d,37 ℃条件下放置1 d病毒完全失活。

     

    Abstract:
    ObjectiveTo observe the stability of severe acute respiratory syrdrome coronavirus (SARS-CoV-2) in cell cultures at different temperatures so as to provide basic data and scientific basis for the research and control of COVID-19 epidemic.
    MethodsThe Vero E6 cells inoculated with SARS-CoV-2. According to TCID50, SARS-CoV-2 with different dilution (10-1, 10-3, 10-5, 10-6)were stored at 37 °C, 22.5 °C, and 4 °C for one to seven days, and then infectious titer was determined by micro cytopathogenic effect assay, observing cytopathic effect (CPE), and real-time fluorescence quantitative testing.
    ResultsSARS-CoV-2 was stable under 4 °C. The infectivity of high concentration (10-1 dilution) under 22.5 °C for seven days gradually decreased, while lower concentration completely lost infectivity after one day. The virus lost infectivity when stored at 37 °C for more than one day.
    ConclusionSARS-CoV-2 is highly stable at 4 °C, sensitive to heat, and related to virus concentration.
