Influence of sample pre-treatment on PCR results of salmonella in food

  • Objective To compare the different effects on DNA detection of two kinds of specimen pre-treatment methods in food salmonella detection.Methods We analyzed the influence of pre-treatment on salmonella DNA detection, using static and blending sampling method to deal with kinds of food. Meanwhile we detected the influence on salmonella DNA detection by cleaning or not cleaning Enrichment Broth centrifugal sedimentation.Results It was found to be more accurate for the result to use static method to deal with cooked food, aquatic products, raw, dairy than blending method, and the difference found in the result was statistically significant. In the comparison of the outcome from cleaning and not cleaning the food Enrichment Broth centrifugal sedimentation, it was found to be more accurate for the outcome to clean the Enrichment Broth centrifugal sedimentation of cooked food, raw meat, dairy, but aquatic products were not affected by the treatment.Conclusion We should use supernatant fluid instead of blending fluid, when conducting salmonella DNA detection of food Enrichment Broth, and clean the Enrichment Broth centrifugal sedimentation, in order to achieve more accurate PCR results.
