高效液相色谱-串联质谱法测定蛋黄中4种类胡萝卜素着色剂的含量

Simultaneous determination of 4 carotenoid pigments in yolk by liquid chromatography-tandem mass spectrometry

    目的建立液相色谱-串联质谱法同时测定蛋黄中4种类胡萝卜素着色剂(角黄素、柑橘皮类胡萝卜素、β-阿朴-8′-胡萝卜素酸乙酯和β-阿朴-8′-胡萝卜素醛)的方法。
    方法蛋黄经乙腈提取,采用Waters Xterra MS C18色谱柱分离;质谱采用电喷雾正离子电离模式和多反应扫描监测模式进行测定,以外标法定量。
    结果4种类胡萝卜素着色剂的检出限均为2.5 ng/g;在2.5~200 ng/g范围内线性关系良好,相关系数r>0.99;回收率在82.24%~109.2%之间,相对标准偏差为1.0%~3.0%(n=6)。
    结论本方法灵敏度高、准确性好,适用于蛋黄中4种类胡萝卜素着色剂的检测。

     

    Abstract:
    ObjectiveTo establish a method for simultaneous determination of four carotenoid pigments (canzthaxanthin, citranaxanthin, β-apo-8-carotenoic acid ethyl ester, β-apo-8-carotenal) in yolk by liquid chromatography-tandem mass spectrometry (LC/MS/MS).
    MethodsYolk was extracted by acetonitrile, and then separated by Waters Xterra MS C18 column.Mass spectrometry used electrospray ionization positive ionization mode(ESI+) and multiple reaction monitoring(MRM) for determination with external standard method for quantification.
    ResultsThe detection limit of the 4 carotenoid pigments were 2.5 ng/g.The method showed a good linearity over the range of 2.5~200 ng/g with r > 0.99.The average recoveries were in the range of 82.24%~109.2%, while the relative standard deviations (RSD) were 1.0%~3.0% (n=6).
    ConclusionThe established method is highly sensitive and accurate, which is suitable for the detection of four carotenoid pigments in yolk.
