Effects of exercise on the celluar immune function of mice induced by PM2.5

    目的 本研究旨在探讨运动对细颗粒物(PM2.5)所致小鼠细胞免疫功能改变的影响。
    方法 将24只雄性C57BL/6小鼠随机分为过滤空气组(n=6)、浓缩PM2.5组(n=6)、运动后暴露过滤空气组(n=6)和运动后暴露浓缩PM2.5组(n=6)。运动组小鼠每天在小鼠跑步机上运动1 h后, 休息1 h, 然后运动组和非运动组小鼠均暴露于浓缩PM2.5或过滤空气。运动暴露24周后, 使用流式细胞仪技术检测小鼠脾脏CD3+CD4+CD8a+T细胞、CD18+T细胞和CD154+T细胞的数量。
    结果 暴露仓、对照仓和室外空气中PM2.5浓度的中位数分别为49.24、12.18、32.25 μg/m3。PM2.5暴露导致CD3+CD4+CD8a+T细胞数量增加, CD154+T细胞数量降低(P < 0.05), 而跑步运动明显降低CD3+CD4+CD8a+T细胞数量(P < 0.05), 提高CD154+T细胞数量(P < 0.05)。
    结论 暴露破坏机体免疫系统稳态, 有规律的跑步运动能够维持免疫系统平衡, 增强机体对PM2.5的抵抗能力。

     

    Abstract:
    Objective To explore the effect of exercise on the celluar immune function of mice induced by PM2.5.
    Methods Twenty-four male C57BL/6 mice were randomly assigned into four groups, filtered air, concentrated PM2.5, exercise and filtered air, and exercise and concentrated PM2.5, with six mice in each group.The mice in the exercise group ran on the treadmill for 1 h every day, and then the mice in the four groups were respectively exposed to concentrated PM2.5 or filtered air after 1 h of rest.After twenty-four weeks, flow cytometric assay was used to detect the quantity of CD3+CD4+CD8a+ T cells, CD18+ T cells and CD154+ T cells in spleen in mice of the four groups.
    Results The median concentration of PM2.5 in the exposure chamber, filtered chamber and ambient air were 49.24, 12.18, 32.25 μg/m3, respectively.PM2.5 exposure led to increase in CD3+CD4+CD8a+ T cells (P < 0.05) and decrease in CD154+ T cells (P < 0.05), whereas running was beneficial for decreasing CD3+CD4+CD8a+ T cells (P < 0.05) and increasing CD154+ T cells (P < 0.05).
    Conclusions PM2.5 exposure induces perturbation of the immune system.Regular running proves to be helpful for maintaining the balance of the immune system and improving the body′s resistance to PM2.5.
