酵母多糖致老年SIRS大鼠模型多器官功能及炎症因子变化的研究

Multiple organ function and inflammatory factor change induced by zymosan in aged SIRS rat models

  • 摘要: 目的 研究酵母多糖(Zym)致老年全身炎症反应综合征(SIRS)大鼠模型的多器官功能及炎症因子变化。方法 壮年和老年SD大鼠,均采用腹腔注射Zym-石蜡混悬液制备SIRS模型。按照不同注射剂量分为750 mg/kg模型组、500 mg/kg模型组、250 mg/kg模型组、125 mg/kg模型组,同时设立空白对照组。观察各组大鼠造模前后的体温、呼吸频率、白细胞计数、心率以确定造模成功。检测空白对照组及各组造模后动脉血气分析指标:pH、二氧化碳分压(PaCO2)、氧分压(PaO2)、血氧饱和度(SaO2),外周血液检测生化指标:丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、肌酐(Cr),以及肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)。计量资料分析采用t检验。结果 成年500 mg/kg组和250 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组壮年大鼠比较差异有统计学意义(P<0.01,P<0.05);壮年125 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组壮年大鼠比较,差异无统计学意义(P>0.05)。老年大鼠500 mg/kg组,250 mg/kg组,125 mg/kg组血气分析指标、血清学检测指标、炎症因子与空白对照组老年大鼠比较差异有统计学意义(P<0.01,P<0.01,P<0.05);老年大鼠与壮年大鼠各相同造模剂量组(500 mg/kg组,250 mg/kg组,125 mg/kg组)血气分析指标、血清学检测指标、炎症因子比较差异有统计学意义(P均<0.01)。结论 腹腔注射Zym-石蜡混悬液制备壮年大鼠SIRS模型以500 mg/kg为佳,老年大鼠SIRS模型以250 mg/kg为佳。相同剂量Zym老年组各器官功能比壮年组差,炎症因子升高。

     

    Abstract: Objective To investigate the changes of multiple organ function and inflammatory factors in the aged rats with systemic inflammatory response syndrome (SIRS) model induced by zymosan.Methods By intraperitoneal injection of Zym-paraffin suspension were prepared SIRS models of adult and aged SD rats. According to different injected doses were set up groups of 750 mg/kg, 500 mg/kg, 250 mg/kg, 125 mg/kg model group, and a blank control group. The success of the model building was determined by observation on the body temperature,respiratory rate,white blood cell count, and heart rate before and after the model was built. Arterial blood gas analysis was made in blank control group and model groups, including:potential of hydtrogen(pH),partial pressure of carbon dioxide(PaCO2),partial pressure of oxygen(PaO2), oxygen saturation(SaO2),and peripheral blood biochemical indexes were detected, including:alanine aminotransferase(ALT),aspartate aminotransferase (AST),creatine kinase(CK),lactate dehydrogenase(LDH),creatinine(Cr), and tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10). Measurement data analysis was done by using t test.Results As the adult 500 mg/kg group and 250 mg/kg group were compared with the adult blank control group,the differences found between them in the results of blood gas analysis, serum test indexes and inflammatory factors were statistically significant (P<0.01,P<0.05). The results of blood gas analysis, serum test index and inflammatory factor in adult 125 mg/kg group were similar to those in adult blank control group and there were no statistically significant differences(P>0.05). As the aged 500 mg/kg group,250 mg/kg group,125 mg/kg groupand were compared with the aged blank control group,the differences found between in the results of blood gas analysis, serum test index and inflammatory factor were statistically significant (P<0.01,P<0.01,P<0.05).As the aged rats were compared with the samedose adult rats (500 mg/kg group, 250 mg/kg group, 125 mg/kg group),the differences found between them in the results of blood gas analysis, serum test index and inflammatory factor were statistically significant(P<0.01,P<0.01,P<0.01).Conclusion The SIRS model of 500 mg/kg in adult rats with intraperitoneal injection of Zym-paraffin suspension was better than models of other dosage, and the dose of 250 mg/kg preferred for the SIRS model of the aged rats. With the same dose of zymosan, the function of each organ in the elderly group were not as good as that in the adult rats,and the levels of inflammatory factors in the elderly group were higher.
